Classification. Retatrutide (developmental code LY3437943) is a synthetic single-molecule peptide designed to act on three receptors: the glucose-dependent insulinotropic polypeptide (GIP) receptor, the glucagon-like peptide-1 (GLP-1) receptor and the glucagon receptor. For this reason it is commonly described in the literature as a "triple agonist."

How it has been studied. Retatrutide was developed by Eli Lilly and Company and has been evaluated in receptor-pharmacology work, preclinical models and company-sponsored clinical trials, with phase 2 results published in peer-reviewed journals including The New England Journal of Medicine. Researchers typically compare it with single- and dual-receptor agonists to understand how activity at multiple incretin and glucagon receptors differs.
